Arabidopsis enhancer-trap lines
Figure GUS staining in the enhancer-trap Arabidopsis lines. We are using the enhancer-trap lines created by the research group of Dr. Tomas Jack, Department of Biological Sciences Dartmouth College. In this system, the vector pD991 which is the derivative of the binary plant transformation vector pCGN1547 is used. The vector contains -60 CaMV minimal promoter fused to uidA (GUS) gene. When the T-DNA insertion containing the minimal promoter-GUS construct is integrated, enhancer elements in genomic DNA adjacent to the T-DNA insertion induces GUS expression by the minimal promoters. We are characterizing the GUS expression specifically in the embryo (A, C) and the endosperm (B, D) of germinating Arabidopsis seeds.
